Perlozzo's professional playing career began when he was drafted by the Minnesota Twins in the 18th round of the 1972 MLB Draft. As an infielder, he spent several years in the minor leagues, playing for various teams within the Twins' organization. His playing career also included stints with the San Diego Padres and the Seattle Mariners organizations. Although he never made it to the major leagues as a player, his time on the field provided him with a deep understanding of the game, which would serve him well in his subsequent roles.

Transitioning from player to coach, Perlozzo found his niche in coaching and management. His coaching career began in earnest with the New York Mets organization, where he served as a minor league manager. His leadership and knowledge of the game quickly became apparent, and he moved up the ranks to become a major league coach.

Perlozzo's major league coaching career included positions with several teams. He served as a third base coach for the New York Mets from 1987 to 1989, before moving on to the Cincinnati Reds, where he held the same position from 1990 to 1992. His reputation as a skilled coach continued to grow, leading to a role with the Seattle Mariners as their third base coach from 1993 to 1995.

In 1996, Perlozzo joined the Baltimore Orioles as their third base coach, a position he held for several years. His tenure with the Orioles was marked by his ability to work effectively with players and his strategic acumen. In 2001, he was promoted to bench coach, further solidifying his role within the team's coaching staff.

The pinnacle of Perlozzo's coaching career came in 2005 when he was named the interim manager of the Baltimore Orioles following the resignation of Lee Mazzilli. Later that year, he was appointed as the team's full-time manager. Perlozzo's managerial stint with the Orioles lasted until June 2007. During his time as manager, he faced the challenges of leading a team in a highly competitive division, striving to bring out the best in his players and improve the team's performance.

After his managerial tenure with the Orioles, Perlozzo continued his coaching career with the Philadelphia Phillies. He joined the Phillies as their third base coach in 2009, contributing to the team's success and helping them reach the World Series that year. His experience and expertise were valuable assets to the Phillies' coaching staff.
